تثبیت آنزیم اورهآز روی نانو بیوفیبریلهای آمیلوئیدی حاصل از آلبومین سرم گاوی
آنزیم اورهآز (EC.3.5.1.5) از دسته هیدرولازها است که هیدرولیز اوره را به آمونیاک و دی اکسیدکربن کاتالیز میکند. این آنزیم کاربردهای مختلفی در متابولیسم نیتروژن، تهیه واکسن، کیتهای تشخیص اوره، صنایع نوشابهسازی و غیره دارد. در این تحقیق از نانورشتههای آمیلوئیدی حاصل از آلبومین سرم گاوی بهعنوان بستری جدید برای تثبیت آنزیم اورهآز استفاده شد. تولید نانو رشتههای آمیلوئیدی با سه تکنیک طیفسنجی کنگورد، اسپکتروفلوریمتری و اسپکتروپلاریمتری بهینهسازی شده و رشتههای حاصله با تصاویر میکروسکوپ الکترونی مورد تأیید قرار گرفتند. سپس آنزیم اورهآز با استفاده از مولکولهای گلوتارآلدهید و با ایجاد پلهای عرضی روی رشتههای آمیلوئیدی تثبیتشده و فاکتورهای سینتیکی آن با آنزیم آزاد مقایسه شدند. بیشترین میزان تولید رشتههای آمیلوئیدی پس از 48 ساعت انکوباسیون آلبومین سرم گاوی با غلظت 10 میلیگرم بر میلیلیتر و دمای 70 درجه سانتیگراد در بافر سیترات-فسفات با pH برابر 4 بهدست آمد. آنزیم تثبیتشده نسبت به شکل آزاد دارای قابلیت نگهداری و پایداری بیشتری بود و فعالیت ویژه بالاتر و نیز Km کوچکتری را نشان داد. دمای بهینه از 40 به 70 درجه سانتیگراد و pH بهینه نیز از 6 تا 7 به 6 تا 9 در آنزیم تثبیتشده بهبود یافت. بنابراین رشتههای آمیلوئیدی با داشتن گروههای شیمیایی مختلف، برای تثبیت آنزیم اورهآز مناسب بودهاند. بهبود ویژگیهای سینتیکی و پایداری آنزیم اورهآز در اثر تثبیت بر رشتههای آمیلوئیدی امکان استفاده گستردهتر از این آنزیم را در صنایع مرتبط فراهم میآورد.

اثر عوامل آگونیست و آنتاگونیست گیرنده های دوپامین بر ترشح بزاق در موش صحرایی
ارتباط دوجانبه بین هسته شکمی میانی هیپوتالاموس و سیستم دوپامینی، ممکن است نقش مهمی در ترشح غدد برون ریز دستگاه گوارشی، رفتارها و عادات تغذیه ای ایجاد نماید. هدف از این مطالعه بررسی نقش گیرنده های دوپامینی در تنظیم غدد برون ریز و آیا ترشح بزاق تحت سیستم عصبی مرکزی است یا فقط در اثر تغییرات سطح گرسنگی، تشنگی و هضم مواد غذایی در دهان صورت می پذیرد؟ 90 سر موش صحرایی تحت بیهوشی عمومی زیر دستگاه استرئوتاکس قرار گرفتند و به 9 گروه: دو گروه کنترل، شم a و b، گروه SCH23390 (آنتاگونیست گیرنده دوپامینی D1) گروه بروموکریپتین (آگونیست گیرنده های دوپامینی D1 و D2)، گروه مخلوط SCH23390 با بروموکریپتین، گروه پیلوکارپین و گروه مخلوط پیلوکارپین و بروموکریپتین تقسیم شدند. میزان ترشح بزاق برحسب حجم (میلی لیتر) اندازه گیری و با کمک برنامه SPSS گروه ها با هم دیگر مقایسه شدند (تست Anova). یافته های مطالعه نشان داد که گروه SCH23390 و گروه مخلوط به طور معنی داری حجم بزاق را افزایش می دهند. بروموکریپتین به تنهایی ترشح پایه بزاق را تغییر نداد. پیلوکارپین ترشح بزاق را افزایش داد و بروموکریپتین در مخلوط با پیلوکارپین بطور معنی داری میزان ترشح بزاق را کاهش داد. SCH23390 بتنهایی و در حضور بروموکریپتین بطور معنی داری ترشح بزاق را افزایش داد. یافته های پژوهش حاضر نشان میدهند که سیستم دوپامینی هسته شکمی میانی هیپوتالاموس روی ترشح بزاق تاثیر بسزایی دارد به صورتی که آگونیست های دوپامین میزان آن را کاهش و آنتاگونیست های دوپامین میزان ترشح بزاق را افزایش میدهند.

بررسی اثر نانو ذرات آهن بر تغییرات هیستوپاتولوژیک تخمدان در موشهای تیمار شده با ایزونیازید
هدف از مطالعه حاضر بررسی اثر نانوذرات آهن بر تغییرات هیستوپاتولوژیک تخمدان در موش های تیمار شده با ایزونیازید میباشد. در این تحقیق 50 سر موش صحرایی ماده بالغ به طور تصادفی به 5 گروه 8 تایی تقسیم شدند؛ گروه کنترل (بدون تیمار)، گروه شاهد (روزانه mg/kg2/0 سرم فیزیولوژی)، گروه تجربی 1 (mg/kg50 داروی ایزونیازید به صورت خوراکی)، گروه تجربی 2 و 3 (mg/kg50 داروی ایزونیازید به صورت خوراکی و به ترتیب به همراه mg/kg150 و mg/kg250 نانوذرات آهن به صورت درون صفاقی) دریافت کردند. نانوذرات 12 روز متوالی تزریق گردید. بعد از 24 ساعت، نانوذرات ذر ترکیب با ایزونیازید تزریق گردید. پس از گذشت 15 روز موش های تمام گروهها تشریح و تخمدان آن ها جهت مطالعه خارج و در فرمالین 10% قرار داده شدند. نتایج نشان می دهد، در گروه دریافتکننده ایزونیازید کاهش فولیکول بدوی، اولیه (تک لایه و چند لایه)، ثانویه، گراف و جسم زرد مشاهده شد (05/0>P). در گروه دریافتکننده ایزونیازید و نانواکسید آهن (150) تعداد فولیکول بدوی، ثانویه و گراف دارای افزایش معنادار و در گروه ایزونیازید و نانواکسید آهن (250) تعداد فولیکول بدوی، اولیه تک لایه، ثانویه و گراف دارای افزایش معناداری نسبت به گروه ایزونیازید از خود نشان میدهند (05/0>P). با توجه به مطالب ذکر شده، ایزونیازید به علت خاصیت اکسیدانی سبب تخریب بافت تخمدان در گروههای دریافتکننده خود شده است و نانواکسید آهن به واسطه خاصیت آنتی اکسیدانی خود تا حدودی این عوارض را کاهش داده است.

مطالعه فعالیت آنتی اکسیدانی اسانس آویشن بر روی هپاتوتوکسیسیتی ناشی از نانو ذره آهن
در این مطالعه به بررسی اثرات حفاظتی اسانس آویشن شیرازی در برابر آسیب کبدی ناشی از نانو ذرات آهن در رت های نر بالغ پرداخته شده است. حیوانات به سه گروه تقسیم شدند: گروه کنترل منفی؛ حیوانات به مدت سه روز نرمال سالین دریافت کردند . گروه کنترل مثبت؛ mg/kg b.w 200 نانو ذره آهن به صورت درون صفاقی به رت های نر به مدت سه روز تزریق شد. گروه تیمار با اسانس آویشن شیرازی؛ حیوانات به مدت 3 روز mg/kg b.w 200 و 100 اسانس آویشن را به صورت تزریق درون صفاقی دریافت کرده اند. پس از 3 روز از قلب خونگیری و بافت کبد به منظور بررسی پاتولوژیکی و بیوشیمی برداشته شد. فعالیت کبدی با استفاده از پارامترهای بیوشیمیایی مختلف مانند آلانین ترانسفراز (ALT)، آسپارتات آمینو ترانسفراز (AST) ، آلکالین فسفاتاز (ALP)، ظرفیت آنتی اکسیدانی کل پلاسما (FRAP)، لیپید پر اکسیداسیون (LP) و گلوتاتیون احیاء (GSH) مورد بررسی قرار گرفت. نتایج این تحقیق نشان داد، که نانو ذرات باعث افزایش AST و LP و کاهش GSH و FRAP می شوند، اما بر روی ALT و ALP اثری ندارد. مصرف اسانس آویشن AST و LP و GSHرا به حالت نرمال بر می گرداند، اما بر روی FRAP تأثیری ندارد. در پایان، تیمار رت ها با نانو ذرات آهن موجب آسیب اکسیداتیو کبدی شده، و استفاده از اسانس آویشن می تواند در جلوگیری و بهبود این آسیب ها موثر باشد.

شناسایی گونهای و بررسی اکولوژیکی خرچنگRhithropanopeus harrrissi (Gould, 1841) (Crustacea: Decapoda) در سواحل دریای خزر
این مطالعه در چهار ایستگاه واقع در سواحل شهرستان نور (دو ایستگاه)، محمود آباد و رویان (استان مازندران) با هدف شناسایی و تعیین برخی از خصوصیات اکولوژیک خرچنگ Rhithropanopeus harrrissi در سواحل دریای خزر انجام گرفت. نمونهها پس از جمعآوری به آزمایشگاه منتقل و با استفاده از کلیدهای شناسایی معتبر شناسایی شدند. بررسی بر روی 192 نمونه خرچنگ نشان داد که آنها متعلق به گونه harrrissi R. هستند. به منظور انجام تأیید شناسایی گونهای این نمونهها با نمونههای متاتیپیک موجود در موزه جانورشناسی دانشگاه تهران مقایسه و مورد تأیید شناسایی گونهای قرار گرفتند. خصوصیات ریخت شناسی، نسبت جنسی، فراوانی و پراکنش این گونه نیز مورد بررسی قرار گرفت. آنالیز واریانس یک طرفه با استفاده از آزمون LSD نشان داد، فراوانی این گونه بین ایستگاه دو (ساحل پارک جنگلی نور) با ایستگاههای یک (ساحل بخش غربی شهر نور) و سه (ساحل محمود آباد) دارای تفاوت معنادار است (05/0P

فون حلزونهای رودخانه جاجرود دراستان تهران طی سالهای 95-1394 مورد بررسی قرار گرفت. مجموعا تعداد 23 گونه حلزون برای اولین بار از رودخانه جاجرود استان تهران شناسایی شد که متعلق به 17 جنس از 19 خانواده است و بر اساس خصوصیات مورفومتری (شکل، اندازه، نقوش روی صدف، اپرکول، دریچه صدف، راستگرد یا چپگرد بودن صدف، محل قرارگیری سوراخ تنفسی ، خط میانه پشتی، موکوس و رنگ آن، اندازه بدن، سوهانک، خط کلیه و دستگاه تولید مثلی) شناسایی شدند. بیشترین و کمترین تنوع گونه ای در میان حلزونهای شناسایی شده به ترتیب با 14 و 8 گونه مربوط به ایستگاههای 2 و 6 از رودخانه جاجرود که به فاصله 36 کیلومتر از یکدیگر قرار دارند، است. بزرگترین خانواده از نظر تنوع گونه ای خانواده Helicida با هشت گونهCochlicella acuta، Euomphalia pisiformis،Helicella krynickii، Helix lucorum lnnaeus، Helicella derbentina، Euomphalia ravergieri، Caucas.tachea atrolabiata، Levantina dschulfensis است. همچنین از بین گونههای شناسایی شده مهمترین گونه از لحاظ پزشکی در انتقال بیماریهای انگلی در انسان و دام گونههای Lymnaea truncatula است. گونههای Limax maculates 8Arion ater نیز برای اولین بار از ایران گزارش می گردد.

مطالعه فونستیکی راسته جوندگان (Order: Rodentia) منطقه مرند، شمال غربی ایران
جوندگان بزرگترین راسته رده پستانداران را تشکیل میدهند و ازنظر پزشکی و کشاورزی دارای اهمیت زیادی میباشند. طی مطالعات فونستیکی روی گونه های مختلف راسته جوندگان در منطقه مرند (استان آذربایجانشرقی) در شمال غرب ایران در سالهای 1391 تا 1394، 6 گونه جمعآوری و شناسائی گردید. در این روش نمونهها با استفاده از تلههای زندهگیر و مردهگیر و طعمه مسموم از مراتع، باغات، خانههای روستائی و اطراف شهر جمعآوری و بر اساس خصوصیات مورفولوژیکی گونههای: 1- Mus musculus، 2- Ellobius fuscocapillus، 3-Microtus socialis،4-Apodemus sylvaticus ، 5-Meriones persicus،6- Cricetulus migratorius شناسائی شدند. کلید شناسائی و اشکال جهت شناسائی آنها در این منطقه فراهم گردید.

واگرایی ریختی اشکول (Glis glis Linnaeus, 1766) در امتداد جنگلهای هیرکانی شمال ایران، شاهدی بر وجود خردپناهگاه در طی آخرین دوره یخبندان
ردپای نوسان های اقلیمی و پیش روی و پس روی یخچال ها در اعصار گذشته را می توان در برخی از جانداران مشاهده نمود. تغییر گسترۀ پراکنش به سمت عرض های جنوبی در دورۀ پیش روی یخچال ها و بقای این جانداران در پناهگاه ها و انتشار مجدد به عرض های جغرافیایی بالاتر، باعث ایجاد تغییراتی ریختی و ژنتیکی شده و با بررسی این تغیرات می توان گسترۀ حضور پناهگاههای مهم به ویژه در دوره پلیستوسن را مشخص نمود. در این پژوهش شواهدی اولیه در خصوص نقش خردپناهگاهی محدودۀ شرقی جنگلهای هیرکانی کشور در آخرین دوره یخبندان بزرگ ارائه می گردد. در این راستا مطالعه، گونۀ اشکول (Glis glis Linnaeus, 1766) که دارای ویژگی های بوم شناختی و زیستی نسبتا منحصر به فردی میباشد به عنوان گونه مدل انتخاب گردید. استخوان فک زیرین افراد متعلق به جمعیتهای مختلف در امتداد جنگلهای خزری ایران به شیوه استاندارد تصویربرداری شده و تحلیل های ریخت سنجی هندسی انجام شد. نتایج حاصل از این تحلیلها با نتایج به دست آمده از مطالعات ژنتیکی مقایسه گردید. نتایج حاکی از واگرایی ریختی شدید جمعیت واقع در منتهی الیه بخش شرقی پناهگاه هیرکانی بر اساس صفات فک زیرین میباشد که کاملا منطبق بر یافته های ژنتیکی پیشین میباشد. همچنین ویژگی های ریخت شناختی ظاهری از قبیل میانگین وزن، میانگین طول بدن و طول دم نیز اختلاف معناداری را با سایر جمعیتهای مورد بررسی در مرکز و منتهی الیه غربی محدوده جنگلهای خزری نشان م

تأثیر جنستئین بر روی برخی پارامترهای بیوشیمیایی و آنزیمی پلاسمای اسپرمی مولدین نر ماهی قرمز
در این مطالعه اثرات جنستئین و 17بتا-استرادیول بر برخی پارامترهای بیوشیمیایی و آنزیمی پلاسمای سمینال ماهی قرمز در مرحله اسپرمیشن مورد بررسی قرار گرفت. ماهیان مولد نر (با وزن تقریبیg 30 تا g40) تحت تزریق داخل عضلانی g/gµ 5 (G5) و g/gµ50 جنستئین (G50)، g/g µ10 هورمون 17بتا-استرادیول، g/gµ10روغن ذرت + DMSO قرار گرفتند. سپس از ماهیان اسپرمگیری به عمل آمد و برخی ترکیبات غیرآلی ، ترکیبات آلی و آنزیمی پلاسمای سمینال اندازهگیری شد. نتایج نشان داد که در ترکیبات غیرآلی، ترکیبات آلی و آنزیمی پلاسمای سمینال بین تیمارهای مختلف اختلاف معنیداری وجود دارد (05/0P ). بیشترین غلظت سدیم، کلسیم و منیزیم به ترتیب در تیمارهای G50 و E2 مشاهده شد. غلظت فسفر و کراتینین در تیمار E2 افزایش یافت، در صورتی که غلظت اوره و تریگلیسرید پلاسمای سمینال در تیمارهای E2، G5 و G50 نسبت به گروه شاهد کاهش یافت. همچنین کاهش معنیداری در غلظتهای ALT و AST در تیمارهای E2، G5 و G50 نسبت به گروه شاهد مشاهده شد. در صورتی که غلظت ALP در تیمارهای E2 و G50 به طور معنیداری افزایش یافت. نتایج این پژوهش بر این نکته تاکید دارد که میزان جنستئین به عنوان یک ترکیب فیتواستروژنی در جیره غذایی مولدین باید بررسی گردد چرا که حضور این ترکیبات استروژنی در دوزهای بالا، از طریق تغییر در ترکیبات پلاسمای سمینال باعث تغییر معنیداری در کیفیت اسپرم خواهد شد.

بررسی فون، توزیع و حفاظت سوسماران استان البرز
چکیده استان البرز با وسعتی برابر با 5833 در غرب استان تهران واقع شده است. اطلاعات اندکی درباره خزندگان و توزیع آنها در استان البرز در دسترس است. در مطالعه حاضرکه در سال های 95-1394 در استان البرز صورت گرفته، فون سوسماران این استان مورد شناسایی قرار گرفت و نقشه توزیع سوسماران شناسایی شده، تهیه شد. نتایج حاصل از این مطالعه نشان داد که 13 گونه سوسمار متعلق به خانوادههای آگامیده، لاسرتیده، جکونیده و سنسیده به قرار زیر Paralaudakia caucasia، Trapelus agilis، Phrynocephalus persicus، Darevskia defilippi، Lacerta strigata، Eremias persica ، Eremias fasciata، Eremias papenfussi، Ophisops elegans، Mesalina watsonana، Bunopus crassicaudus، Ablepharus bivittatus و Eumeces schneideri در این استان زیست میکنند. در مطالعه حاضر دو رکورد جدید از توزیع سوسماران ثبت شد گونه Lacerta strigata در ارتفاعی 900 متر بالاتر از ارتفاع ثبت شده قبلی در شمال استان یافت شد، همچنین غربیترین رکورد توزیع Eremias fasciata در این استان ثبت شد که از رکورد قبلی آن در مرکز ایران تا 200 کیلومتر فاصله دارد.

مناطق کلیدی جهت حفاظت از خرس قهوهای (Ursus arctos Linnaeus, 1758) ماده در البرز مرکزی
هدف از این مطالعه رسیدن به یک مدل مطلوبیت زیستگاهی مبتنی بر دادههای حضور خرس قهوهای ماده و شناسایی فاکتورهای مهم در انتخاب زیستگاه می باشد. شناسایی این مناطق نقش بسیار مهمی در حفاظت از خرس قهوهای خواهد داشت. در این راه با توجه به عوامل مؤثر در تعیین کیفیت زیستگاه، نتایج حاصل بهصورت یک نقشه پیشبینی پراکنش زیستگاههای مطلوب و اولویتبندی دامنه تغییرات متغیرهای زیست محیطی تأثیرگذار بر این مطلوبیت ارائه میشود. لذا دادههای لازم برای مدیریت و حفاظت گونه و زیستگاه مطلوب آن از طریق تعیین پاسخ گونه به محیطزیست و پارامترهای شناسایی شده تسهیل خواهد شد. در این بررسی از روش بیشترین بینظمی برای مدلسازی زیستگاه خرس قهوهای در منطقه حفاظت شده البرز مرکزی استفاده شد. دادههای در دسترس برای تعیین اولویت انتخاب زیستگاه شامل یک مجموعه نقاط حضور گونه براساس مشاهدات میدانی و همچنین متغیرهای زیست محیطی منطقه مورد مطالعه است. صحت مدل به دست آمده از نتایج، حد استاندارد بالای 89/0 را نشان میدهد. براساس تحلیل جک نایف نیز متغیر بارندگی سالیانه مؤثرترین پارامترهای حضور گونه بوده و همچنین پوشش گیاهی منحصر به فردترین اطلاعات را در اختیار مدل قرار داده است.

مطالعه مقدماتی زیستشناسی وزغ خراسانی Bufotes oblongus (Nikolskii, 1896) در شرق ایران
این تحقیق به منظور مطالعه برخی از ویژگیهای زیستی وزغ خراسانی Bufotes oblongus، از ماه فروردین 1391 تا تیر ماه 1392، در قنات قصبه شهر گناباد درخراسان رضوی و سد و چشمه فصلی روستای مهوید و قناتهای کوران و بلده شهر فردوس و قنات محمدآباد شهر قاین در خراسان جنوبی انجام گرفت. لاروهای این گونه، در طول روز با کمک تور دستی با چشمه یک میلیمتر و تمامی نمونههای بالغ، در شب با کمک دست جمعآوری شدند. مشخصه مهم این گونه، داشتن برآمدگی رو به پایین غده پاروتوئید درعقب پرده صماخ و همچنین کوچک بودن قطر پرده صماخ است. بیشترین و کمترین طول بدست آمده از این گونه در زیستگاه طبیعیاش، 27/8 سانتیمتر (جنس ماده) و 59/5 سانتیمتر (جنس نر) بود. نسبت طول ساق پا به اندازه بدن برای این گونه و در این تحقیق، 35/0 و نسبت طول غده پاروتوئید به اندازه بدن، 21/0 به دست آمد. نتایج این تحقیق در ارتباط با بررسی یک چشمه فصلی، نشان داد که آلودگی آب آبگیرها به علت ورود مدفوع حیوانات اهلی به داخل محل تخمریزی و تکوین لاروها، استفاده از این چشمه به عنوان آبشخور دامها و همچنین تبخیر آب به علت گرمای هوا، جزء عوامل تهدیدکننده حیات این گونه به شمار میآیند. به نظر میرسد برنامهریزی و انجام پروژههای آموزشی به منظور بالا بردن آگاهی مردم نسبت به نقش ارزنده دوزیستان، میتواند راهکاری ارزشمند برای حفاظت این رده از مهرهداران، به ویژه در مناطق نیمه بیابانی کشور باشد.

بررسی تأثیر رویدادهای زمینشناسی خلیج فارس بر توزیع و پراکنش گکوی سنگلاخ بلانفورد (Pristurus rupestris) با استفاده از ژن میتوکندریایی 12S
خانواده Sphaerodactylidae گکوهایی هستند که در امریکای شمالی و جنوبی، کارائیب، جنوب اروپا، شمال افریقا، خاورمیانه و آسیای مرکزی توزیع شدهاند. گکوهای خانواده Sphaerodactylidae جنس Pristurus شامل 23 تا 26 گونه میباشند. گکوی سنگلاخ بلانفورد Pristurus rupestris یکی از کوچکترین گونههاست که در شرق عربستان و جنوب ایران پراکنده شده است. در این مطالعه منشا و نحوه گسترش این گونه در ایران و جزایری که حضور این گکو در آنجا تایید شده است، بررسی گردید. همچنین تأثیرات نوسانهای آب و هوایی بر روی پراکنش این گونه در طی دورههای زمینشناسی با استفاده از نشانگر میتوکندریایی (12S) مورد مطالعه قرار گرفت. از اینرو 10 نمونه از محدوده پراکنش این گونه در ایران جمعآوری و به همراه نمونههای موجود در بانک ژن مورد بررسی قرار گرفت. نتایج نشان دادند که نمونههای ایران متعلق به کلاد شرق (P. rupestris) هستند. همچنین نمونههای متعلق به جزایر ایرانی خلیج فارس به جز جزیره خارک تفاوت چندانی با سرزمین اصلی نشان ندادند. شبکه هاپلوتایپی نشان داد که نمونههای ایران به همراه نمونههای عمان در یک هاپلوگروپ قرار گرفته و هاپلوتایپ مرکزی نیز مربوط به عمان میباشد. از اینرو احتمال میرود که نمونههای ایران در طی دوره پلیستوسن از جمعیت سواحل عربی مشتق شدهاند و در زمان خشک بودن خلیج فارس به ایران رسیدهاند. با توجه به اینکه موانع ژئومورفولوژیکی بر روی الگوی پراکنش خزندگان تأثیر مستقیم میگذارند آنها میتوانند شاخص خوبی برای بررسی الگوهای جغرافیای تبارشناختی باشند.
